Calcium Flux Assay

Calcium flux assays are primarily used to study changes in intracellular Ca²⁺ concentrations. Calcium ions play a key role as second messengers in many biological processes. To illuminate the role of these ions, calcium flux assays typically use fluorescent probes that change their fluorescence properties upon binding to Ca²⁺, allowing changes in intracellular Ca²⁺ concentrations to be monitored by techniques such as fluorescence microscopy and flow cytometry. Calcium flux assays are often used to screen drugs for G protein-coupled receptors (GPCRs) and calcium channels.

cAMP Assay

The cAMP flux assay focuses on changes in intracellular cAMP levels. cAMP is an important secondary messenger that regulates a variety of cellular functions. cAMP flux assay methods include ELISA, bioluminescence, and high-throughput homogeneous ELISA, which provide high sensitivity and high-throughput detection. In addition, live cell imaging techniques are used to monitor changes in cAMP levels in real time, such as the use of FRET sensors for dynamic measurement of cAMP levels.

The combination of calcium flux and cAMP assays offers a more comprehensive approach to the study of signaling. For example, in certain cell types, there is a sophisticated interplay between calcium and cAMP signaling that may influence the cell's response to diverse stimuli.
