APOE Antibodies

Apolipoprotein E (APOE) is a glycoprotein that is involved in the regulation of lipid metabolism. In the central nervous system, it is found in astrocytes, microglia, blood vessel wall cells, and choroid plexus cells. APOE is polymorphic and contains three allelic mutants: APOE*ε2, APOE*ε3, and APOE*ε4.

APOE Overview

Research indicates that APOE is a significant risk factor for late-onset Alzheimer's disease (AD). APOE*ε2 protects against AD, while APOE*ε4 raises the risk. The number of APOE genotypes carried also influences the likelihood of acquiring AD. The risk of AD increases 3-4 times in individuals carrying one APOE*ε4, but increases 9–15 times in those carrying two. APOE modulates the expression of brain β-amyl plaques, tau protein, and TDP43 protein in AD patients, as well as normal brain function.

The Influence of APOE on AD Pathogenesis

  • (Amyloid Beta) Aβ metabolism: APOE plays a role in the formation of β-amyloid plaques in the brain by regulating amyloid protein production, clearance, and deposition near neurons. Research indicates that APOE*ε4 can significantly increase the production and accumulation of Aβ in neurons.
  • Neuroinflammation: APOE can affect inflammation in the brain, and the APOE ε4 allele may promote the development of AD by modulating the inflammatory response of microglia.
  • Cerebrovascular: APOE impairs the integrity and function of cerebral blood vessels. The APOE ε4 allele may damage cerebral blood vessels and accelerate the onset of AD.
  • Synaptic function: The APOE ε4 allele may modify synaptic signaling and plasticity, leading to AD pathogenesis.
