Stain Kits
Researchers employ histological staining to analyze how cells, structures, and molecules are arranged in tissues and organs. In this technique, the tissue of interest is preserved in a chemical fixative and sliced, or cut into very thin sections. Various staining techniques are then used to provide contrast to these seemingly uniform sections. In neuroanatomy, histological techniques are often used to observe and study the tissues of the nervous system.

Nissl Stain
The Nissl stain was introduced in the late 19th century by the German neurologist Franz Nissl. Nissl used a class of basic dyes that stained the nuclei of all cells and the clumps of material around the nuclei of neurons, called Nissl bodies. Nissl stain has the following advantages: it can distinguish neurons from glial cells; it allows histologists to study the arrangement or cytoarchitecture.

Golgi Stain
However, not all problems can be solved by Nissl stain. Italian histologist Camillo Golgi invented a new way to learn more about neurons. In 1873, Golgi discovered that soaking brain tissue in a solution of silver chromate would cause a small portion of the neurons to turn completely black. This method is now called the Golgi stain. This revealed that the cell body of the neuron (i.e., the area around the neuron nucleus shown by Nissl staining) is only a small part of the entire neuron structure.
