TDP-43 Antibodies

TDP-43 (TAR DNA binding protein 43), expressed by the TARDBP gene, can bind both DNA and RNA and plays a critical role in intracellular RNA transcription, alternative splicing, and regulation of mRNA stability. Amyotrophic lateral sclerosis (ALS) and frontotemporal lobar degeneration with TDP-43 inclusions (FTLD-TDP) are both defined by lesions caused by aberrant TDP-43 aggregation. TDP-43 is the third protein associated with Alzheimer's disease (AD), and it plays a key role in AD progression. Apart from tau and amyloid-β (Aβ), most AD patients also have TDP-43 accumulation in the limbic regions. This accumulation has been dubbed the limbic predominance of age-related TDP-43 neuropathological changes in encephalopathy (LATE-NC).

The Impact of TDP-43 on AD Pathology

The specific mechanism of TDP-43 in AD is still not completely clear. TDP-43 can influence a wide range of physiological processes by forming complex core molecules and controlling gene transcription. However, in AD, TDP-43 frequently loses its normal function and forms toxic aggregates in neurons. This aberrant buildup of TDP-43 is thought to promote neuronal death, leading to clinical symptoms of AD such as memory loss and cognitive decline.

Furthermore, researchers have discovered a link between the TDP-43 protein and disease progression. AD patients with aberrant TDP-43 often proceed more quickly than patients with normal TDP-43. These individuals may acquire clinical signs early and experience rapid AD progression.
