TREM2 Antibodies

Triggering receptor expressed on myeloid cells 2 (TREM2) has been suggested to play a critical role in the etiology of Alzheimer's disease (AD). The TREM2 gene is mainly expressed in myeloid cells and tissue-specific macrophages. Microglia are myeloid cells in the central nervous system that exhibit high levels of TREM2. However, it is controversial if TREM2 is expressed exclusively in a subset or in all microglia in mice and humans.

Structure of TREM2

TREM2 is a transmembrane glycoprotein present on the cell surface that consists of three unique domains.

  • The first is an extracellular sensor protein with an immunoglobulin domain that recognizes pathogen-associated molecular pattern (PAMP) molecules, damage-associated molecular pattern (DAMP) molecules, cellular debris, lipids, and lipid proteins. Moreover, the TREM2 extracellular domain responds to accumulating Aβ in AD. Cleavage activation by ADAM family enzymes ADAM10 and ADAM17 results in the production of a secreted form of the TREM2 extracellular domain (sTREM2), which may protect microglial survival and proinflammatory signal propagation.
  • The second TREM2 domain, the transmembrane domain, is connected with the intracellular adaptor protein DAP (DNAX activating protein) 12. Pro-inflammatory cytokine production and release, as well as phagocytosis and debris clearance, are all likely regulated by the interaction between TREM2 and DAP12. The timing and intensity of microglial activation directly influence neuronal survival and function, and changes in TREM2/DAP12 function have a direct effect on this.
  • TREM2's third domain is its short cytoplasmic tail (TREM2 ICD), which is released by γ-secretase cleavage. The function of the TREM2 ICD is unknown.
