DAPI firmly binds to the A-T-rich region in DNA to form a fluorescent complex. When combined with DNA, it preferentially stains ds-DNA and has a high quantum yield (φf = 0.92).

Application Notes
It is preferred for staining dead cells. DAPI is less effective as a staining agent for living cells because it cannot effectively pass through membranes in living cells. Therefore, it may be necessary to use higher concentrations.
DAPI must penetrate and/or fix the cell to enter the cell and bind to DNA.
Because DAPI emits blue light, yellow fluorescence, green fluorescent molecules (such as fluorescein and GFP) or red fluorescent colors (such as Texas Red) there is almost no fluorescence overlap. Therefore, it is convenient for multiple determination.
DAPI has many applications, but it is usually used for cell imaging, cell counting, cell sorting (based on DNA content), apoptosis analysis and HCA (high content analysis).
