Native calcium GPCR detection provides a fast and reliable method for detecting changes in intracellular calcium. Cells expressing target receptors that signal through calcium are preloaded with calcium-sensitive dyes and then treated with compounds. When stimulated, the receptor signals the release of intracellular calcium, which leads to an increase in dye fluorescence. This assay provides a homogeneous assay format for detecting calcium mobilization and measuring intracellular calcium, providing valuable information about GPCR and ion channel activation status. The signal is measured on a fluorescence plate reader equipped with a fluid handling device, which can detect rapid changes in fluorescence during complex stimuli. These assays are designed to be compatible with 96-well and 384-well plate formats.
