Description

Human periodontal ligament fibroblasts are isolated from periodontal ligament tissue; periodontal ligament (periodontal ligament, periodontal space) is composed of dense connective tissue. Most fibers are arranged in bundles, one end of the fibers is buried in the cementum, and the other end is buried in the bone wall of the alveolar socket, so that the teeth are fixed in the alveolar socket; there are nerves, blood vessels, lymphatic and epithelial cells in the periodontal ligament Wait. Fibroblasts are the main cellular components of loose connective tissue, differentiated from mesenchymal cells during embryonic stage. Fibroblasts are larger, with clear outlines, mostly protruding spindle-shaped or star-shaped flat structures, with regular oval nuclei and large and conspicuous nucleoli. Fibroblasts have vigorous functional activities, weak basophilic cytoplasm, and obvious protein synthesis and secretion activities. Under certain conditions, they can achieve mutual transformation with fibroblasts; fibroblasts are resistant to different degrees of cell degeneration, necrosis and tissue. Defect repair plays a very important role. The newly isolated periodontal ligament fibroblasts were round and had good refractive index, and were suspended in the medium. After 30 minutes, the cells adhered to the wall, and some of them began to protrude pseudopodia, which appeared as small protrusions; after 6 hours, the cells were basically completely adherent, stretched into a spindle shape, and the nuclei were clear and evenly distributed. Rapidly, 5-7 days, the cells are in a confluent state, the cells are closely arranged, and some overlap and grow, flat, with large cell bodies, transparent cytoplasm, and large nuclei, which are oval and light in color. Cells fuse and connect to each other in a network; cells are flattened in the shape of a protruding spindle or star. Periodontal ligament fibroblasts (PLFs), as the main cells of periodontal ligament, are the main interstitial cells of periodontal ligament. It also participates in the process of disease, repair and regeneration of periodontal tissue. Now, the use of periodontal ligament cells to establish in vitro models has become an important means for researchers to study periodontal diseases.

The in vitro culture period of primary cells is limited. It is recommended to use the special growth medium provided by Creative Biolabs and the correct handeling method to culture the cells, so as to ensure the best culture state of the cells.
