Description
Human neural progenitor cells (NPC) are derived from induced pluripotent stem (iPS) cells produced using STEMCCA Cre excisionable constitutive polycistronic (OKSM) lentivirus. The cell-permeable TAT-Cre recombinase was used to remove viral transgenes, and the genomic DNA extracted from human iPSC cells by quantitative PCR was excised.

These hiPSC-derived NPCs proliferate into adherent cell monolayers, and the expression of appropriate neural stem cell markers including Nestin and Sox-2 exceeds 80%. The cells are provided at the 3rd generation and can be expanded 3 to 5 times using Estem ™ Neural Expansion Medium (NCC200529ZP). By using Human ES / iPS Neuronal Differentiation Medium (NCC200538ZP) or Estem ™ Neuronal Differentiation Medium (NCC200541ZP), human NPC can be further differentiated into terminal neurons. Differentiation results in more than 80-90% of cells mainly expressing neuronal markers bIII-tubulin and MAP2.

Human iPSC-derived NPCs can be used in a variety of research applications, including neurotoxicity studies, co-culture applications, and screening molecules that induce or inhibit preferential differentiation into mature neurons.

Handling Advice
Frozen cells:Upon receipt, frozen ampoules should be transferred directly to gaseous phase liquid nitrogen without delay, unless they are to be used straight away.

Growing cells: Growing cell cultures should be checked on receipt using an inverted microscope. Immediately check the cell density upon arrival for any obvious defects. If the cell density is too high (more than 80% confluent) subculture the cells (harvest and reseed) immediately.

Quality Control
Mycoplasma was tested for sterility, post-freezing viability, and short-term repeat (STR) analysis for cell line identification. Cytochrome oxidase I (COI) was analyzed for cell line species identification.
