Description

SV40 was used to immortalize the N27 rat dopaminergic neuron cell line, which was isolated from E12 rat midbrain tissue. The neurological deficiency improved over time after the N27 cell line was introduced into the striatum of 6-hydroxydopamine-damaged rats (an animal model of PD). For both in vitro and in vivo research, this immortalized cell line can be employed as a model for dopaminergic neurons because it has been carefully characterized in dopamine biosynthesis and neurotoxicity investigations.

The cell line has been screened using the membrane filtration testing methods to confirm the absence of aerobic, anaerobic and fungi microorganisms.

Handling Advice

Frozen cells:Upon receipt, frozen ampoules should be transferred directly to gaseous phase liquid nitrogen without delay, unless they are to be used straight away.

Growing cells: Growing cell cultures should be checked on receipt using an inverted microscope. Immediately check the cell density upon arrival for any obvious defects. If the cell density is too high (more than 80% confluent) subculture the cells (harvest and reseed) immediately.

Warnings

Store under recommended storage conditions (liquid nitrogen). Do not expose to high temperature. After expiration, discard all remaining reagents. It is recommended to use cells within ten generations.

Quality Control

Mycoplasma was tested for sterility, post-freezing viability, and short-term repeat (STR) analysis for cell line identification. Cytochrome oxidase I (COI) was analyzed for cell line species identification.
