Description

N'-nicotinamide methyltransferase (NNMT) (EC 2.1.1.1) uses S-adenosylmethionine (SAM) as a donor to catalyze the N-methylation of nicotinamide, pyridine and other analogs, thereby Produce 1-methylnicotinamide (MNA). NNMT plays an important role in the regulation of metabolic pathways and is expressed at high levels in a variety of cancers, neurodegenerative diseases, obesity and diabetes, indicating that it is a potential molecular target for treatment. Our NNMT inhibitor screening kit uses SAM as the methyl donor and nicotinamide as the substrate. NNMT methylates nicotinamide to produce S-adenosyl homocysteine ​​(SAH) and 1-methyl nicotinamide. SAH is hydrolyzed by SAH hydrolase to form homocysteine, and its free sulfhydryl group is detected by Thiol detection probe. The enhanced fluorescence signal generated can be measured at Ex / Em = 392/482 nm. In the presence of NNMT inhibitors, enzyme activity is inhibited, resulting in a decrease in fluorescence. The detection kit is a simple, sensitive and rapid tool that can be used to screen potential NNMT inhibitors.
