Description

Human anti-Ki67 antibody recognizes Ki67 cell cycle related proteins. Ki67 is expressed in proliferating cells, but not in resting cells. The expression of this antigen preferentially occurs in the G1, S, G2, and M late stages of the cell cycle, while the antigen cannot be detected in cells in the G0 phase. Therefore, Ki-67 antigen expression is used in tumor pathology to detect proliferating cells in neoplastic diseases. In cultured cells, Ki-67 is expressed in the nucleoli of interphase cells. The Ki67 gene contains 15 exons. Exon 13 encodes the Ki67 repeat region of the Ki67 motif with 22 amino acids. The shorter isoform lacks exon 7. Northern blot analysis showed that multiple transcripts in proliferating cells ranged from about 8.9 to 12.5 kb, but not in resting cells. Western blot analysis showed the expression of 320 and 359 kDa proteins. Antisense oligonucleotides inhibited cell proliferation in a dose-dependent manner, indicating that Ki67 protein expression may be an absolute requirement for cell proliferation. In the cell, Ki67 is mainly located in the G1 phase of the perinucleolus region. In the later stage, it can also be detected in the entire nucleus, mainly in the nuclear matrix. In mitosis, it is present on all chromosomes.
