Product Overview


Rabies virus (RABV) is an enveloped, bullet-shaped, negative single-stranded RNA virus of the genus Rabies virus of the Rhabdoviridae family. The RNA genome of the virus is approximately 12 kb, which encodes 5 viral proteins, including nucleoprotein (N), Phosphoprotein (P), matrix protein (M), glycoprotein (G) and viral RNA polymerase (L) from the 3'end to the 5'end.

Through reverse genetics, a recombinant RABV constructed by Wickersham et al. Based on the SAD-B19 vaccine strain. The glycoprotein gene of modified RABV is completely removed (G deletion) and loses the ability to cross synapses, but its replication and transcription are not affected. RABV-∆G can be used to reverse high-brightness and finely labeled neurons. If we provide G protein first (inject AAV-G first), then the recombinant RABV will have the ability to propagate retrograde in the synapse.

In order to further improve the targeting effect of G-deficient RABV on specific starting cells, the virus was modified by binding to the envelope protein EnvA of avian sarcoma and leukemia virus (ASLV). The infectivity of these viruses is limited to cells that carry TVA receptors. Of course, the receptor TVA and EnvA proteins must be provided first.
