Description

Brain uptake of therapeutic antibodies is severely limited by their size. To achieve enhanced BBB crossing, Creative Biolabs developed a BBB shuttle antibody platform by utilizing the endogenous macromolecule transportation pathway, known as receptor-mediated transcytosis (RMT). The engineered antibody-based carrier is believed to significantly to increase the macromolecule brain entry to combat CNS diseases.

Figure 1 shows the GAL-Elk1 luciferase assay in rat L6 cells. Cells were co-transfected with expression vectors for the indicated FGFR isoforms together with GAL-Elk1, SV40-Renilla luciferase and Gal-responsive firefly luciferase reporter genes.

Figure 2 shows food intake (left), body weight (middle) and blood glucose (right) in db/db mice after a single intraperitoneal injection of R1Mab2 or control IgG at a dose of 3 mpk on day 0. Control mice were pair fed (PF) with adjusted food intake until day 11. On day 11, the food intake of R1MAb2-treated mice returned to normal, so all mice were fed ad libitum after day 11 (AL). N=7~12. p<0.001.

Figure 3 shows a GAL-Elk based luciferase assay in HEK293 cells expressing FGFRlc to compare R1MAb2 and a DNA mutant of R1MAb2 (R1MAb2 DANA).
