Introduction to Antisense Oligonucleotide (ASO)
Antisense oligonucleotides (ASOs) usually refer to chemically modified short-chain nucleic acids with about 15-25 nucleotides. Their sequences are complementary to the sense sequences of target mRNA according to the principle of Watson-Crick base pairing. ASOs are widely used for gene silencing and thus serve as an important tool for gene function study. In traditional drug discovery workflows, ASOs were commonly used for target identification and validation. What's more, ASOs are now being explored as therapeutics themselves. In theory, any RNA molecule in the transcriptome can be inhibited by nucleic acid-based drugs. Once inside the cell, the highly target-specific nucleic acid drugs can discriminate single nucleotide mismatches. Compared with small molecule drugs, nucleic acid drugs exhibit a faster development speed.

Antisense Oligonucleotide (ASO) Screening Service
ASO-based therapy is an emerging field of drug development against a variety of gene-specific diseases. By using single-stranded oligonucleotides, RNA expression and splicing can be altered to reduce, restore, or modify protein expression. Through the identification of different mechanisms of action, such as RNaseH recruitment, splicing modifications, and miRNA targeting, personalized antisense therapy could be successfully predicted. Creative Biolabs can perform in vitro ASO screening using various disease cellular models, such as human primary cells and iPSC-derived neurons. For an identified RNA target, the ASO screening strategy allows the discovery of the optimal ASO.
