ELISA

Figure 1 shows the ELISA response of 1E16 mAb and 115 mAb using intact Nav1.7 DII VSD. Data are shown as mean SEM (n=3).

1080 mAb produced about 26% inhibition of Nav1.7 current at 25 μg/ml.

Inhib

Figure 2 shows that voltage sensor targeting 1E16 inhibits human Nav1.7 in HEK293 cells.

Representative current traces from HEK293 cells expressing hNav1.7 in the absence or presence of 1 iuM 115 (A) and 100 nM 1E16 (C). The current-voltage relationship in the absence (o) or presence (D) of 1 iuM 115 (B) and 100 nM 1E16 (D) was generated using a 30 ms voltage step between -80 and +60 mV, from hold Start the potential at -120 mV in 10 mV increments.

Inhib

Figure 3 shows that 1E16 inhibits Nav1.7 in a subtype-specific and state-dependent manner in HEK293 cells.

State (usage)-dependent inhibition of human Nav1.7 by 1E16.

FuncS

Figure 4 shows the effect of 1E16 mAb on Nav1.7, caused by a specific interaction between the tip (ring) of the voltage sensor paddle and 1E16.

From HEK293 cells expressing human Nav1.7 channel (A), in the presence of 1E16 (100 nM) and peptide (1 μM) (B) and in the presence of 1E16 (100 nM) only (C).

FuncS

Figure 5 shows the effect of mAbs on sustained sodium currents (INaP) in small neurons from the entire dorsal root ganglion (DRG) of naive and nerve-injured mice (CCI.).

From HEK293 cells expressing human Nav1.7 channel (A), in the presence of 1E16 (100 nM) and peptide (1 μM) (B) and in the presence of 1E16 (100 nM) only (C).
