Figure 1 shows the binding of anti-TNC A2 antibody 2B10 as wild type (wt) and glycoengineered (ge) versions to TNC A2 on U87MG cells.
Cells were incubated with indicated concentrations of mu18H10, HTA 125 (commercially available anti-human TLR4 non-blocking monoclonal antibody) or antibody control followed by incubation with LPS (15 ng/ml). IL-8 levels were assessed 16 hours after LPS treatment.
Figure 2 shows the binding of 2B10-derived human IgG to TNC A2 expressed on U87MG glioblastoma tumor cells as determined by flow cytometry.
Cells were incubated with indicated concentrations of mu18H10, HTA 125 (commercially available anti-human TLR4 non-blocking monoclonal antibody) or antibody control followed by incubation with LPS (15 ng/ml). IL-8 levels were assessed 16 hours after LPS treatment.
Figure 3 shows immunohistochemical images of human tissue stained with 2B10-mouse IgG at 100× (left panel) and 400× (middle panel) magnifications.
Staining with an isotype control antibody is shown at 100X magnification (right panel). Top: normal tissue, bottom: tumor tissue from the brain.
Figure 4 shows the SPR-based kinetic analysis of the binding of 2B10 anti-TNC A2 Fab fragment to human TNC A2.
Staining with an isotype control antibody is shown at 100X magnification (right panel). Top: normal tissue, bottom: tumor tissue from the brain.
Figure 5 shows the SPR-based kinetic analysis of 2B10 anti-TNC A2 Fab fragments binding cynomolgus monkey TNC A3.
Staining with an isotype control antibody is shown at 100X magnification (right panel). Top: normal tissue, bottom: tumor tissue from the brain.
Figure 6 shows the SPR-based kinetic analysis of the binding of the 2B10 anti-TNC A2 Fab fragment to murine TNC A3.
Staining with an isotype control antibody is shown at 100X magnification (right panel). Top: normal tissue, bottom: tumor tissue from the brain.
