Product Overview

Description

Cell line information: DOR-tGFP_U2OS
Official Full Name: Delta Opioid receptor (DOR)
DNA Accesion Number: Gene Bank NM_000911
Host Cell: U2OS
Resistance: G418
Storage: Liquid Nitrogen

Features

The DOR-tGFP U2OS cell line contains 3 million U2OS cells, which stably express the human Delta opioid receptor (DOR) labeled with tGFP protein at the N-terminal. The Creative Biolabs DOR redistribution cell line is designed to detect potential agonists/antagonists against DOR and regulate its activation and subsequent redistribution processes within the cell. This cell line will allow image analysis of compound-induced stimuli. In high content analysis (HCA), the use of Deltorphin II as a DOR agonist has validated this highly reproducible assay.

Assay

Delta Opioid Receptor Internalization Assay

Assay Description

The Delta opioid receptor internalization analysis from Creative Biolabs allows the detection of compounds or their ability to modulate Delta opioid receptor (OPRD1) activation and the following redistribution processes within the cell. When the ligand binds to OPRD1, it activates G Protein, which is internalized in large, high-strength vesicles.

The assay has measured fluorescence OPRD1 redistribution by increasing the fluorescence around the nucleus using image analysis algorithms. The high reproducibility of this assay allows monitoring of the delta opioid receptor redistribution process in high-throughput screening.

Mycoplasma Testing

The cell line has been screened using the luciferase based mycoplasma detection kit to confirm the absence of mycoplasma species.

Sterility Testing

Sterility testing was performed in accordance with USP and EP regulations. All of our sterility testing is performed in an isolator or clean room environments. The cell line has been screened using the membrane filtration testing methods to confirm the absence of aerobic, anaerobic and fungi microorganisms.

Genetic Stability Testing

Cell genetic stability study was perfomed under ICH guidelines. We provide guidance on the appropriate testing program upon your requirements.
