Figure 1 depicts the binding of murine monoclonal antibody DF200 to the common de 5 terminus of various human KIR2DL receptors.
Figure 2 depicts that mouse monoclonal antibody DF200 neutralizes KIR2DL1-mediated inhibition of NK cell cytotoxicity against Cw4-expressing target cells, shown at an effector/target (E/T) ratio of 4/1.
Figure 3 depicts the neutralization of KIR2DL1-mediated cytotoxic inhibition by murine monoclonal antibody DF200, the Fab fragment of DF200, and conventional murine antibodies specific for KIR2DL1 (mAb EB6 and XA-141) or KIR2DL2/3 (mAb GL183 and Y249)15 Inhibition of Cw4-positive target cells, and inhibition of KIR2DL2/3-mediated NK cell cytotoxicity against Cw3-positive target cells.
Figure 4 depicts the reconstitution of lysis of 20 HLA-Cw4 positive target cells by NK cells expressing KIR2DL1 in the presence of DF200 and the F(ab')2 fragment of the EB6 antibody.
Figure 5 depicts the induction of NK-mediated killing by cross-reactive murine (DF200 and NKVSF1 (Pan2D)) and human (1-7F9, 1-4F1, 1-6F5, and 1-6F1) mAbs,
Figure 6 depicts the induction of NK-mediated killing by the same antibodies as in Figure 5 . mAb induced (reconstituted) killing of KIR2DL1-expressing NK cells (YTS 30 KIR2DL1) by B-EBV cells expressing endogenous HLA-Cw4. E/T ratio = 2.30 pg/ml mAb.
Figure 7 depicts the results of FACS analysis.
