Features

The Creative Biolabs SSTR3 Internalization Assay cell line contains U2OS cells, which stably express human somatostatin receptor type 3 labeled with tGFP protein at the N-terminal. The Creative Biolabs SSTR3 internalization cell line is designed to detect potential agonists/antagonists against SSTR3 and regulate its activation and subsequent intracellular redistribution process. This cell line will allow image analysis of compound-induced stimuli. In high content analysis (HCA), the use of somatostatin 28 as the SSTR3 agonist validated this highly reproducible assay.

Assay Description

The SST3 somatostatin receptor internalization analysis from Creative Biolabs allows the detection of compounds or their ability to modulate SST3 somatostatin receptor (SSTR3) activation and the following redistribution processes within the cell. When the ligand binds to SSTR3, It activates G protein, which is internalized in large, high-strength vesicles.

The assay has measured fluorescence SSTR3 redistribution by using image analysis algorithms by increasing the fluorescence around the nucleus. The high reproducibility of this assay allows monitoring of the SST3 somatostatin receptor redistribution process in high-throughput screening.
