Figure 1 shows the histogram of the FACS analysis detailing the binding of various IGF-1R-specific antibodies to IGF-1R.
Figure 2 depicts Western blot analysis of NIH 3T3 IGF-1R+ membrane extracts and recombinant extracellular domain of IGF-1R and IR probed with mAb 12B1 after SDS-PAGE electrophoresis under non-reducing and reducing conditions.
Figure 3 shows the 125IIGF-1 binding inhibition experiment. Plot total specific IIGF-1 binding (%) as a function of ligand concentration on a semi-log graph. Specific binding values are the mean of experiments performed in triplicate.
Figure 4 shows the results of immunohistochemical studies (IHC) of various cell lines using a control antibody (IgG) and 12B1. The data show that 12B1 differentially stains membranes of various cell lines.
Figure 5 Upper panel: Goat polyclonal IGF-1R IHC (@ 1.0 μg/ml) in colon cancer. 20x (left) and 40x (right - higher magnification image of inset) objectives. Tumor cells have variable membrane staining. Bottom: 12B1 monoclonal IGF-1R IHC (@ 0.75 μg/ml) in colon cancer.
Figure 6 Left panel: Goat Poly IGF-1R IHC (@ 1.0 μg/ml) in colon cancer #2. 40× objective lens.
Tumor cells have plasma membrane staining. Right: 12B1 monoclonal IGF-1R (@ 0.75 μg/ml) in colon cancer #2 in the same tumor area shown on the left. 40× objective lens. Plasma membrane and apical globular, Golgi-like staining were present in tumor cells.
Figure 7 Top left: Goat polyclonal IGF-1R IHC (@ 2.0 μg/ml) in normal skin #3. 40× objective lens. Staining of the epithelial cell membrane on the outer side of the hair follicle (light/thin arrow).
