Glutamate carboxypeptidase II [GCPII, EC 3.4.17.21; N-acetylated α-linked acid dipeptidase (NAALADase), prostate specific membrane antigen (PSMA) or folate hydrolase (FOLH1)] is one A kind of zinc metal peptidase, multifunctional protein. In humans, it is mainly expressed in the nervous system, prostate, small intestine and kidney. In men, PSMA is overexpressed in patients with prostate cancer, therefore, it is used as a diagnostic marker for the disease. The brain isoenzyme NAALADase plays an important role in the biosynthesis of glutamate and can be used to treat neuronal damage caused by excessive glutamate in the brain. Our PicoProbe™ Glutamate Carboxypeptidase II Activity Assay Kit is a simple plate-based fluorescence assay for measuring GCPII activity in biological samples. The provided substrate is transaminated in the presence of GCPII producing glutamate. The detection system is based on an enzymatic reaction, in which the fluorescent probe is reduced and a stable signal is generated. The reduced fluorophore produces a strong signal that is proportional to the amount of active GCPII in the sample. This detection method can detect glutamate carboxypeptidase II as low as 0.5 µU.
