Glutamate carboxypeptidase II [GCPII, EC 3.4.17.21; N-acetylated α-linked acid dipeptidase (NAALADase), prostate specific membrane antigen (PSMA) or folate hydrolase (FOLH1)] is one A multifunctional zinc metal peptidase. In the past decade, GCPII has been studied as a promising pharmacological target for the treatment of various pathological diseases and diseases, and great efforts have been made to discover effective GCPII inhibitors. The brain isoenzyme NAALADase plays an important role in the biosynthesis of glutamate and can be used to treat neuronal damage caused by excessive glutamate in the brain. For example, in animal models with several acute and chronic diseases (stroke, neuropathic and inflammatory pain, schizophrenia, depression, or amyotrophic lateral sclerosis), inhibition of GCPII has shown neuroprotective properties. In men, PSMA is overexpressed in patients with prostate cancer, therefore, it is used as a diagnostic marker for the disease. Our glutamate carboxypeptidase II inhibitor screening kit is an in vitro tool for screening potential human GCPII inhibitors. The provided substrate is transaminated in the presence of GCPII producing glutamate. The detection system is based on an enzymatic reaction, in which the fluorescent probe is reduced and a stable signal is generated. In the presence of 2-PMPA (a potent GCPII inhibitor), the enzyme activity is blocked, resulting in a lower fluorescence signal. The assay kit is a simple plate-based format, which can provide fast and reliable tests for high-throughput screening of GCPII inhibitors.
