Figure 1 shows the histogram of the FACS analysis detailing the binding of various IGF-1R-specific antibodies to IGF-1R.
Figure 2 depicts Western blot analysis of NIH 3T3 IGF-1R+ membrane extracts and recombinant extracellular domain of IGF-1R and IR probed with mAb 12B1 after SDS-PAGE electrophoresis under non-reducing and reducing conditions.
Figure 3 shows 125IIGF-1 binding inhibition experiments. Plot total specific IIGF-1 binding (%) as a function of ligand concentration on a semi-log graph. Specific binding values are the mean of experiments performed in triplicate.
Figure 4 shows the results of immunohistochemical studies (IHC) of various cell lines using a control antibody (IgG) and 12B1. The data showed that 12B1 differentially stained the membranes of various cell lines.

There is plasma membrane staining of tumor cells. Right: 12B1 monoclonal IGF-1R (@ 0.75 μg/ml) in Colon Carcinoma #2 in the same area of tumor shown at left. 40× objective. There is plasma membrane as well as apical globular, golgi-like staining in tumor cells.
