Description

Brain uptake of therapeutic antibodies is severely limited by their size. To achieve enhanced BBB crossing, Creative Biolabs developed a BBB shuttle antibody platform by utilizing the endogenous macromolecule transportation pathway, known as receptor-mediated transcytosis (RMT). The engineered antibody-based carrier is believed to significantly to increase the macromolecule brain entry to combat CNS diseases.

Figure 1 EphA2 antibody inhibits malignant tumor cell growth in vitro. Purified EphA2 antibody was incubated with malignant and benign tumor15 cells in soft agar at 37°C for 7 days.

A549 malignant lung cancer cells were incubated with 10 μg/ml or 2.5 μg/ml of EA2 or EA5 monoclonal antibodies or control (PBS). All antibody amounts used inhibited cell growth in soft agar.

Figure 2 EphA2 antibody EA2 inhibits tumor cell growth in vivo.

MDA-MB-231 breast cancer cells were implanted subcutaneously in athymic mice. After tumors grew to a mean volume of 30 to 100 mm3, mice were administered 6 mg/kg of the indicated antibody or negative control intraperitoneally twice weekly for 3 weeks. Total tumor volume was determined after sacrifice. Negative control is black and EA2 is white.

1x105 control or MCF-7EPhA cells were suspended in soft agar in the presence or absence of tamoxifen (TAM, 1 pM) and EphA2 agonistic antibody (EA2, 10 μg/ml). Note that EA2 increased the sensitivity of MCF-7EIhA2 cells to tamoxifen.

EphA2-F was incubated with and bound to 5 immobilized Ephrin Al-Fe. Labeled EA2 was then incubated with the EphA2-ephrin A1 complex. Unlabeled competitors were incubated with the indicated amounts of EphA2-Ephrin A1-EA2 complexes. Competitors are Ephrin Al-Fc (black) or EA2 (white).
