Description

Glyceraldehyde 3-phosphate dehydrogenase (GAPDH) is a metabolic enzyme responsible for catalyzing a step in the glycolytic pathway, which is the reversible oxidative phosphorylation of glyceraldehyde 3-phosphate to glyceraldehyde diphosphate.. Because GAPDH protein is expressed in large quantities and is always required for important "housekeeping" functions, the level of GAPDH mRNA is often measured and used as a standard for other mRNA expression studies. Similarly, a specific antibody against GAPDH is used to measure the expression of GAPDH, which can be used as a protein standard in Western blot experiments to compare the level of this protein with other proteins in cells or tissues. In addition to its role in glycolysis, GAPDH may also have other roles, such as transcription activation (1), and bind to a variety of other proteins, including amyloid precursor protein, huntingtin's polyglutamine bundle ( 2,3). This protein may also play a role in the regulation of apoptosis. Interestingly, when a cell is apoptotic, it migrates from the cytoplasm to the nucleus (4). The control of this is mediated by the NO-mediated S-nitrosylation of GAPDH in the cytoplasm, which then binds and stabilizes the Siah1 E3 ubiquitin ligase, which enters the nucleus together with the nitrosylated GAPDH and initiates specific The protein degradation event caused the nuclear cell to activate the death pathway (5). Recently, GAPDH has been shown to be a substrate of 5'-AMP-dependent protein kinases, causing cellular GAPDH to redistribute to the plasma membrane and inhibit intracellular transport, which is a component of the cell starvation response (6).
