Description

This unique fluorophore has the highest quantum yield of any calcium indicator on the market and can provide the largest signal window in a calcium determination kit. The substantial increase in signal amplitude helps to reignite the low signal screen; including endogenous, primary or stem cell targets. In addition, this new dye is not affected by organic anion exchange proteins, so it can be measured with almost no probenecid.

When used in quench-free (QF) formulations, this dye is very suitable for quenching sensitive targets or assays that require multiple calcium flux and luminescence measurements in one well.

The FLIPR calcium assay kit provides a homogeneous solution for evaluating GPCRs and ion channels
The FLIPR calcium assay kit utilizes calcium-sensitive dyes that are absorbed into the cytoplasm of cells during incubation. After the ligand-receptor binds, it releases intracellular calcium and binds to the dye, thereby increasing the fluorescence intensity. All FLIPR calcium assay kits use extracellular masking technology to block background fluorescence and increase the signal window of the assay. The FLIPR Calcium 6-QF Analysis Kit is an exception, which can achieve analytical performance without the use of masking techniques, making it ideal for quenching sensitive applications such as multiplexing with luminescent indicators.

The new fluorophore of the FLIPR Calcium 6 detection kit shows the highest signal-to-noise ratio when performing no-wash detection (the initial medium is removed, replaced with detection buffer, and the dye is loaded without washing). Obtain the same high quality performance with or without masking dye.

The FLIPR calcium 5 analysis kit and the FLIPR Tetra system were used to evaluate the antagonistic effect of EC80 acetylcholine on calcium flux in CHO M1 cells, demonstrating a larger signal window and performance consistent with previously published data.
