Product Overview

Description

The kit can use a fluorescent microplate reader to detect the content of glutathione in mammalian cells, thereby detecting early apoptosis events. Decreased cellular glutathione (GSH) is an important antioxidant in mammalian cells. Due to GSH efflux, it occurs in the early stages of mitochondrial-related apoptosis. This kit uses monochlorodiphenyldiamine (MCB), a cell-permeating dye with high affinity for GSH, to detect cellular GSH levels in apoptotic events. The unreacted dye hardly fluoresces. After the dye reacts with GSH, it is catalyzed by endogenous glutathione S-transferase (GST) enzyme, and the dye emits blue fluorescence, which is excited at 394 nm and emits at 490 nm. The intensity of the fluorescent signal generated by the measurement reflects the amount of GSH present in the cell.

Components

Explorer Kit
(1) vial of MCB (10 mM, 100 µL)
Each kit contains enough material for two (2) 96-well or 384-well microplates.

Bulk Kit - PN
(1) vial of MCB (10 mM, 500 µL)
Each kit contains enough material for ten (10) 96-well or 384-well microplates.
Data

Fig.1 Representative data from LinearMax microplate reader
Use data collected from HeLa cells. (A) Live/Dead: Cell viability measured by green (live)/red (dead) fluorescence ratio; (B) Caspase-3/7 R110: Apoptosis measured by caspase 3/7 activity; (C) Valley Glutathione: Apoptosis measured by glutathione content; D&E: Caspase-3/7 NucView488. The SmartTox Caspase-3 / 7 NucView 488 detection kit is also suitable for fluorescence imaging, because the nuclei of intact apoptotic cells are labeled according to their caspase activity. (D) Assess caspase 3/7 activity on a microplate reader; (E) Assess caspase 3/7 using fluorescence imaging.
