qPCR is used to determine changes in steady-state mRNA levels of gene expression in multiple samples, which are usually normalized to the relative expression of internal control genes. Gene-specific primers can amplify target sequences in a cDNA library reverse-transcribed from mRNA, and, like TaqMan® technology, hybridized sequence-specific probes can provide fluorescence from the 5'exonuclease activity of Taq DNA polymerase signal. The accumulation rate of the fluorescent signal is used to quantify the sample cDNA, thereby quantifying the amount of mRNA in the original cell lysate.
The qPCR array contains validated primers and probes for the detection of 90 genes whose expression is related to undifferentiated hPSC or its derivatives at the early stage of differentiation, and six endogenous (housekeeping) control genes. Provide TBP (TATA Box Binding Protein) qPCR control template as a positive control for synthetic DNA.
