Description

TDP43 was originally identified as a protein that binds to the "transactivation response" (TAR) sequence in the long terminal repeat of the HIV-1 viral genome (1). UV cross-linking of HeLa cell extracts revealed a 43kDa protein, which was cloned and sequenced, and revealed two central copies of the RNA recognition motif or RRM domain containing approximately 90 amino acids. RRM is an acronym for RNA Recognition Motif. This domain is found in many proteins that bind single-stranded RNA and some proteins that bind single-stranded DNA. It also has a single binuclear localization sequence in the N-terminal region before the first RRM domain, while the C-terminal region outside the second RRM domain is rich in glycine, asparagine, and glutamine. TDP43 is usually expressed in the central nervous system of mammals, but it accumulates in nuclear inclusions in the neurons of patients with frontotemporal degeneration (FTLD), amyotrophic lateral sclerosis (ALS), Alzheimer's disease, and other diseases (2-4). Disease-related point mutations in TDP43 are almost always found in the C-terminal region. TDP43 is related to FUS/TLS, EWSR1 and TAF15 in protein sequence, and all four proteins have a tendency, especially when they are mutated, they converge into inclusion bodies and are related to a variety of neurological diseases (5,6).
