PP1 Binding Activity Assay (FP Assay)

Background of Protein Phosphatase 1

Synaptic plasticity is involved in the efficacy of neuronal transmission, which contributes to the underlying biological mechanism of memory and learning. The process of synaptic plasticity is based on the mechanism of phosphorylation and dephosphorylation by protein kinases and protein phosphatases. The balance of phosphorylation and dephosphorylation processes is the key to maintaining synaptic plasticity, while imbalance of protein kinases and protein phosphatases may lead to neuropathological conditions. Therefore, the serine/threonine protein PP1 belonging to protein phosphatases plays an important role in synaptic plasticity and neuronal functions.

PP1 Binding Activity Assays

Creative Biolabs uses high-throughput fluorescence polarization (FP) assay to determine the PP1 binding activity of compounds. We use 384-well Low Dead Volume (LDV) microplates, which enable a large amount of compound screening. Fluorescein isothiocyanate (FITC)-labeled peptides and PP1 are then added for later incubation. FITC-labeled PP1 rotate rapidly in the solution and are excited by polarized light with FP signal. When the compounds bind with PP1 to form a complex, the emitted light becomes polarized as a result of the slow movement of the complex. The results of FP signal alterations can be read and calculated by our imaging and analytical platforms.

Applications

PP1 binding activity assay determined by FP assay can be used to screen compounds acting as PP1 inhibitors for maintaining the balance of protein kinases and protein phosphatases to treat neurological disorders. Our high-throughput technologies enable efficient screening of compounds.
