Product Overview

Description

Oligodendrocytes belong to the family of glial cells in the human central and peripheral nervous system. They are multifunctional cells and are involved in many disabling neurological diseases, including adult-onset demyelinating diseases, leukocyte dystrophy and cerebral palsy.

Creative Biolabs' OPCs are induced oligodendrocyte progenitor cells that are reprogrammed from human fibroblasts using a proprietary reprogramming technology. OPCs cells were cryopreserved at a low passage number. OPCs cells are characterized by standard morphological examination and immunocytochemical methods for known marker proteins (such as O4, PDGFalphaR, NG2 and CNPase), and can be induced to express MBP under the induction of T3 hormone.

Mycoplasma Testing

The cell line has been screened using the luciferase based mycoplasma detection kit to confirm the absence of mycoplasma species.

Sterility Testing

Sterility testing was performed in accordance with USP and EP regulations. All of our sterility testing is performed in an isolator or clean room environments. The cell line has been screened using the membrane filtration testing methods to confirm the absence of aerobic, anaerobic and fungi microorganisms.

Genetic Stability Testing

Cell genetic stability study was perfomed under ICH guidelines. We provide guidance on the appropriate testing program upon your requirements.

Handling Advice

Frozen cells: Upon receipt, frozen ampoules should be transferred directly to gaseous phase liquid nitrogen without delay, unless they are to be used straight away.

Growing cells: Growing cell cultures should be checked on receipt using an inverted microscope. Immediately check the cell density upon arrival for any obvious defects. If the cell density is too high (more than 80% confluent) subculture the cells (harvest and reseed) immediately.

Q&As
Can you please recommend a medium?
The Oligodendrocyte Precursor Cell Growth Medium [CAT#: NCL-21P6-105] is used for culturing OPCs, and the Oligodendrocyte Precursor Cell Differentiation Medium [CAT#: NCL-21P6-106] is used for the OPC differentiation.
Should we seed the cells in a well of a 6-well plate or a T25 flask?
1) Seeding the iOligo cells (post-thaw) into 3 wells of a 6-well plate. Or, seed the iOligo cells into 1 single T25 flask. 2) Please do NOT over-passage the iOligo cells before differentiation. No more than 2X.
What’s the protocol for Matrigel coating (Corning)?
Please see Corning’ s instructions: dilute their Matrigel according to their lot# instructions. Typically, it is ~250µl- 400µl Matrigel in a 25ml solution (DMEM/F-12, for example). Then use 1ml to coat 1 well (6-well plate).
After thawing and centifuging in DMEM/F12. We resuspend in Oligodendrocyte Precursor Cell Growth Medium (no additional factors or Penstrep?)? And then plate in 3 wells of a 6-well plate? 1.5ml of media per well?
The cell culture media is serum-free and ready-to-use. Please do NOT dilute it further with anything! If you are using 6-well plates, each well gets ~1.5mL (instead of 1mL).
How many passages can the cells be cultured without compromising the phenotype of the cells?
We do not recommend more than 2X passages.
What´s the source of the cells?
Human fibroblasts cells.
