Description

This AAV expresses mCherry and TetTox driven by hEf1a promoter in Cre recombinase-dependent manner.
The hEF1a promoter, cloned from the promoter region of human translation elongation factor 1A1 (eukaryotic translation elongation factor 1 alpha 1 (EEF1A1)), is 1.3 kb in length, expresses constitutively, and has no cell tissue specificity.
DIO stands for double-floxed inverse open reading frame. It refers to a genetic arrangement where a transgene of interest is inserted in reverse orientation relative to the 5' promoter and is flanked by oppositely oriented loxP and lox2272 sites. In the absence of Cre expression, the transgene will not be produced. However, in the presence of Cre expression, the transgene will undergo a permanent Cre-mediated recombination or inversion, resulting in the expression of the transgene. This arrangement is known as DIO.
The TetTox gene, short for Tetanus Toxin, encodes a modified tetanus toxin fragment that can selectively inhibit the release of neurotransmitters in the targeted cells.
