Recombinase Tools
Creative Biolabs' recombinase tools are developed using the Cre-LoxP system strategy and can perform knockout, insertion, activation, inversion and translocation at specific sites on DNA. Our recombinase tools are highly efficient and temporally specific, have been validated in both eukaryotes and prokaryotes, and are functional in a variety of tissues and physiological settings. Tissue-specific or inducible promoters can be used to tightly regulate recombinase expression. Recombinase AAVs are excellent tools for a wide range of applications, including gene function research in neurological diseases and gene therapy.
Applications of the Cre-loxP System
- Specific Labeling / Neural Circuit Tracing
Researchers can utilize Cre-strain mice and Cre-dependent viral vectors to mark and modify specific cells. The purpose of specific neural circuit labeling and functional research can be achieved by combining the Cre recombinase system with AAV serotypes (rAAV2/9, rAAV2/retro, and rAAV2/1) when using neural tracing technology to study the composition and function of specific neural circuits in the brain.
- Virus-dependent Gene Recombination
While genetic recombination of transgenic mice is time-consuming, costly, and not highly specific to regions or tissues, an increasing number of researchers are turning to the Cre-loxP system for more flexibility. A common way is to introduce Cre or loxP elements using viruses and then combine them with a transgenic mouse.
Virus-dependent gene recombination provides the following advantages: It can achieve more robust regional and cell-specific gene recombination. The costs of virus preparation, storage, and injection are relatively modest. By delivering viruses to transgenic mice, the experimental time can be significantly reduced.
Other Recombinase Systems
System | Description |
Flp-FRT | The Flp-FRT system, like the Cre-loxP system, induces gene recombination. The most notable distinction between the two systems is that the recombinases (Cre and Flp) have different optimal reaction temperatures. The ideal temperature for Cre recombinase is 37°C, while Flp recombinase is 30°C. |
Dre-Rox | Dre (D6 site-specific DNA recombinase) is a tyrosine recombinase discovered in bacteriophages. It recognizes the 32-bp DNA sequence rox, which consists of two 14-bp reverse palindromic sequences and a 4-bp intermediate spacer region. Specific recombinases include Dre and Cre. Dre recombinase cannot recognize loxp sites, whereas Cre recombinase cannot detect rox sites. |
vCre-vloxp / sCre-sloxp | vCre and sCre are both Cre recombinase homologs, however their homology is relatively low. Vloxp can be recognized especially by vCre. Sloxp can be recognized especially by sCre. |
