Description

Brain uptake of therapeutic antibodies is severely limited by their size. To achieve enhanced BBB crossing, Creative Biolabs developed a BBB shuttle antibody platform by utilizing the endogenous macromolecule transportation pathway, known as receptor-mediated transcytosis (RMT). The engineered antibody-based carrier is believed to significantly to increase the macromolecule brain entry to combat CNS diseases.

Figure 1 is a graph showing the mean fluorescence (MF) intensity exhibited by human monoclonal antibody (mAb) 5H7, human mAb 7H5, or murine mAb 1C6 bound to CXCR3 transfectants. The 100% value of the mean fluorescence intensity for calculating EC50 is the binding intensity of each antibody at 10 μg/ml. The EC50 values for human mAb 5H7, human mAb 7H5 and mouse mAb 1C6 are 0.5 nM, 1.2 nM and 2.3 nM, respectively.

Figure 2 is a graph depicting the results in the absence ("positive") or presence of antibodies directed against human CXCR3 (murine mAb 1C6, human mAb 5E17, subclone 4, elution tubes (1) or (2) or human mAb 5H7, subclone 6, elution tube (1) or (2)). The results show that human monoclonal antibody 5H7 inhibits CXCR3-mediated chemotaxis in response to IP-10, MIG and I-TAC.

Figure 3 is a graph showing the percent inhibition of the chemotaxis of CXCR3 transfectants to IP-10, MIG or I-TAC by human monoclonal antibody 5H7 in an in vitro chemotaxis assay. Human mAb 51-17 has IC50 of 0.7 μg/ml for IP-10, 2.9 μg/ml for MIG and 1.2 μg/ml for I-TAC.

Figure 4 is a graph showing the percent inhibition of the chemotaxis of CXCR3 transfectants to IP-10, MIG or I-TAC by murine monoclonal antibody 1C6 in an in vitro chemotaxis assay. Mouse mAb 1C6 has an IC50 of 0.8 μg/ml against IP-10, 93 μg/ml against MIG and 23.9 μg/ml against I-TAC.
