Description

Microtubules are protein rods with a diameter of 25 nanometers found in most eukaryotic cells and are related to a family of proteins called microtubule-associated proteins (MAP). MAPs play a crucial role in the regulation of microtubule dynamics and interactions in vivo. MAP2 was found to be a high molecular weight MAP with an SDS-PAGE molecular weight of approximately 280kDa (1-3). A single mammalian MAP2 gene can produce two high-molecular-weight proteins of about 280kDa, called MAP2A and MAP2B, and low-molecular-weight forms commonly called MAP2C and MAP2D, which run on a 60-70kDa SDS-PAGE gel. The 60-70kDa form is present in neurons early in development, but was later replaced by a higher molecular weight form (2). The MAP2A and MAP2B forms consist of a long protein sequence that forms tiny filamentous protrusions from the side of the brain's microtubules, so it is called the projection domain. The epitope of the antibody is mapped to the projection domain, so the antibody is specific for MAP2A and MAP2B. This region is one of the prototypes of the "intrinsic unstructured region", which is a widespread type of protein sequence (4). The MAP2 subtype is only expressed in neurons, especially in the perinuclear and dendrites of these cells. Therefore, antibodies against the MAP2 isotype are excellent markers of neuronal dendrites and can be used to identify neurons in cell cultures and slices (eg 5-9).
