RAGE (receptor for advanced glycosylation end products), also known as AGER, is a 35 Dalton transmembrane receptor of the immunoglobulin superfamily, which was first identified by Neeper et al. in 1992. Its name stems from its ability to bind advanced glycation end products (AGE), which mainly includes glycoproteins, whose sugar groups have been non-enzymatically modified by Maillard reaction. Given its inflammatory function in innate immunity and its ability to detect a class of ligands through a common structural motif, RAGE is often called a pattern recognition receptor. RAGE also has at least one other agonistic ligand: high mobility group protein B1 (HMGB1). HMGB1 is an important intracellular DNA binding protein in chromatin remodeling. It can be passively released by necrotic cells or actively secreted by macrophages, natural killer cells and dendritic cells.
