Block

Figure 1 In vitro characterization of 59R5.

The figure shows that antibody 59R5 is able to block ligand-induced Notch2 and Notch3 signaling.

Block

Figure 2 In vitro characterization of 59R5.

The fig shows that 59R5 binds to a similar epitope as 59R1.

Inhib

Figure 3 Notch receptor antibody 59R5 inhibits tumor formation and growth in vivo.

Figure shows in vivo treatment of PE13 breast tumor cells with antibody 59R5.

Inhib

Figure 4 Notch receptor antibody 59R5 inhibits tumor formation and growth in vivo.

Figure shows in vivo treatment of C28 colon cells with antibody 59R5.

FuncS

Figure 6 In vivo treatment of tumors using Notch2/3 antibody 59R15 in combination therapy.

Mice were injected with PN8 pancreatic tumor cells. Tumors were allowed to grow for 33 days until they had reached an average volume of 120 mm3. The animals were treated with gemcitabine at 20 mg/kg once per week for four week in combination with either control Ab (squares). 59R1 (triangles), or 59R5 (circles).

FuncS

Figure 7 In vivo treatment of tumors using Notch2/3 antibody 59R15 in combination therapy.

Mice were injected with PE13 breast tumor cells. Tumors were allowed to grow for 40 days before treatments were initiated. The animals were treated with TAXOL at 15 mg/kg twice per week for 5 weeks, plus either control antibody (squares) or 59R5 (circles). After 5 weeks, the TAXOL treatments were stopped and the antibody treatments continued.

FuncS

Figure 8 Regulation of tumor gene expression after antibody 59R5 treatment.

Figure 8 shows the expression levels of selected genes in stromal cells and selected human genes in PE13 tumor cells following treatment with 59R1, 59R5, or control antibodies.

FuncS

Figure 9. 59R1 reduces PE13 breast cancer stem cell frequency.

Cancer stem cell frequency in PE13 breast tumors after treatment with 59R5 and/or taxol. A single asterisk indicates a statistically significant difference (p<0.05) vs. the control antibody treated group and a double asterisk indicates a significant difference vs. the taxol and control antibody treated group.
