Figure 1 illustrates the PCR synthesis of another human Notch1 immunogenic plasmid N1-NRR-TM(+) cDNA.
Figure 2 illustrates the results of a Notch-1-dependent luciferase reporter assay for two monoclonal antibodies: mAb N248A and mAb-C.
Figure 3 illustrates the change in plasma mAb N248A concentration after a single dose injection of 5 mg/kg into mice. Each data point is calculated based on 3 mice. T 1/2 is the half-life of N248A in mouse serum.
Figure 4 illustrates NICD inhibition in HBP-ALL xenograft tumors following single dose injection of 5 mg/kg in mice. Western blot bands of tumors treated with control antibody D16A were set to 100% intensity, corresponding to 0% inhibition.
The Notch1 mAb, N248A, exhibited potent antitumor activity in this model after 11 days of treatment (i.e., two doses every two weeks).
Figure 5 illustrates growth inhibition of HPB-ALL xenograft tumors by mAb N248A. After tumors grew to 150-300 mm 3 , mice were administered mAbs as indicated. Each group contained 10 mice with random tumor sizes.
Figure 6 PK curves show that N248A mAb reached the maximum concentration (approximately 235 nM) in mouse serum 24 hours after injection. Estimated half-life approximately 4.5 days
