Description

FOS genes and proteins were originally identified as transformation elements in viral oncogenes. The transforming protein is named v-FOS, which stands for viral FOS, and the normal cell non-transforming proto-oncogene is named c-FOS, which stands for cellular FOS. FOS is the acronym for "FBJ Murine Osteosarcoma", and this virus is the first virus in which this gene product has been discovered. The c-FOS protein is a normal gene that acts as an on/off switch that controls the expression of many other genes. The v-FOS form mutates to remain in the on position, which continuously activates other genes and promotes unregulated cell division. Unmutated c-FOS is an "immediate early" gene, so called because protein expression is usually very low, but in response to various stimuli (including serum, growth factors, tumor promoters, cytokines, and ultraviolet light) Will increase radiation rapidly and briefly. The newly expressed c-FOS protein binds to the JUN family and other basic leucine zipper (bZIP) proteins to form multiple activating protein 1 (AP-1) complexes (1). The AP-1 complex specifically activates the expression of many other genes, thereby regulating the cell's response to stimuli, which may lead to cell proliferation, differentiation, tumor transformation, cell apoptosis, and response to stress (2). Therefore, the regulatory expression of c-FOS plays an important role in many cell functions. Site-specific phosphorylation activates c-FOS, and the sumoylation of c-FOS inhibits AP-1 transcriptional activity (3,4). Since c-FOS expression is induced in neurons that rapidly fire action potentials, appropriate c-Fos antibodies can be used to identify activated neurons in the tissue (5). Using current technology, it is possible to track the progress of such cells or obtain data about their mRNA expression.
