Features

The red cAMP Maxensor-ADRβ3 contains U2OS cells stably expressing the red cAMP Maxensor biosensor and adrenergic receptor Beta 3 (unlabeled). Creative Biolabs' cAMP Maxensor-ADRβ3 cell line is designed to test compounds or analyze their ability to modulate the beta 3 adrenergic receptor. When the agonist binds to ADRβ3, the G protein is activated, which in turn triggers a cellular response mediated by cAMP. This cell line has been verified by measuring the increase in cAMP in the cytosol, analyzing the increase in red fluorescence and the distribution of biosensors in the cell. The use of isoproterenol as an agonist in high-throughput screening (HTS) and high-content analysis (HCA) validates this highly reproducible assay.

About Red cAMP Maxensor Biosensor:
The red cAMP Maxensor biosensor is a fluorescent peptide that changes its intracellular location in the presence or absence of cAMP. Before stimulating cAMP production, the fluorescent biosensor is located in the cell membrane. The increase in the concentration of the second messenger leads to changes in the structural folding of the red cAMP Maxensor biosensor, which promotes the relocation of cells in the transport of cell vesicles. In cell lines that co-express the red cAMP Maxensor biosensor and the GPCR of interest, the activity of living cells can be easily quantified by fluorescence granularity image analysis or fluorescence intensity analysis.

Assay Description

β-3 adrenoceptor (β3 adrenoceptor); also known as ADRB3, is a β-adrenoceptor that spans cell membranes. Creative Biolabs' beta3 adrenergic receptor test (ADRB3) allows the use of Maxensor biosensor technology to measure ADRB3 receptor activation. Beta3 adrenergic receptor signaling occurs through adenylate cyclase stimulation through the interaction of trimerized Gs protein, increased cAMP, and downstream L-type calcium channels. The cAMP-Maxensor biosensor measures receptor activation through cAMP accumulation.
