Description

Brain uptake of therapeutic antibodies is severely limited by their size. To achieve enhanced BBB crossing, Creative Biolabs developed a BBB shuttle antibody platform by utilizing the endogenous macromolecule transportation pathway, known as receptor-mediated transcytosis (RMT). The engineered antibody-based carrier is believed to significantly to increase the macromolecule brain entry to combat CNS diseases.

Figure 1 is a graph depicting the effects of 15C11, 2B1, and 1C2 (30 mg/kg) on contextual memory in Tg2576 mice, as determined by contextual fear conditioning assays.

In Tg2576 mice administered 15C11, memory deficit reversal was complete and marked. Asterisks (*) indicate significant differences from wild type and number signs (#) indicate significant differences from vehicle-treated heterozygotes.

Figure 2 is a graph depicting the effect of low doses (3 mg/kg and 10 mg/kg) of 15C11 on contextual memory in Tg2576 mice, as determined by contextual fear conditioning assays.

The data showed no trend for damage with increasing doses of 15C11. The improvement in contextual memory was significant in Tg2576 mice receiving 30 mg/kg of 15C11. Asterisk (*) indicates significant difference from wild type.

Figure 3 depicts the results of the epitope map analysis of 15C11. The results indicate that 15C11 recognizes residues 19-22 (FFAE) of Aβ.

Figure 4 depicts immunoprecipitates of peroxynitrite-treated oligomeric Aβ1-42 preparations precipitated with various Aβ antibodies (3D6, 6C6, 12A11, 12B4, 3A3, 266, 9G8, 15C11, and 6H9) and imaged with 3D6 Western blot. Approximate positions of Aβ1-42 monomer, dimer, trimer, and tetramer bands are shown on the left side of the Figure.
