Product Overview

Description

Brain uptake of therapeutic antibodies is severely limited by their size. To achieve enhanced BBB crossing, Creative Biolabs developed a BBB shuttle antibody platform by utilizing the endogenous macromolecule transportation pathway, known as receptor-mediated transcytosis (RMT). The engineered antibody-based carrier is believed to significantly to increase the macromolecule brain entry to combat CNS diseases.
Notes: The BBB antibody is made-to order and available in a customized format. Please don't hesitate contact us for more details.

Inhib

Figure 1 is a graph showing that antibodies A, A-1, A-2 and A-3 inhibit Notchl as measured using a luciferase reporter assay.

Block

Figure 2 is a graph showing that Antibody A relieves the block of differentiation of mouse C2C12 myocytes caused by activation of Notch signaling.

Block

Figure 3 shows the results of antibody epitope mapping experiments.

Block

Figure 4 is a graph showing that anti-Notchl-NRR antibodies inhibit wild-type and mutant Notchl receptor signaling.

FuncS

Figure 5 is a series of Figures showing that anti-Notch1 NRR and anti-D114 treatment resulted in a significant increase in vessel sprouting and length in the HUVEC (Human Umbilical Vein Endothelial Cell) cell sprouting assay.

FuncS

Figure 6 is a graph showing the effect of anti-Notchl NRR antibodies on tumor growth in an in vivo mouse model. Treatment with anti-Notch1 NRR antibody A-2 prevented tumor growth.

FuncS

Figure 7 is a series of Figures showing examples of intestinal crypts and villi from the small intestine of mice treated with a control antibody (vehicle) or with anti-Notch NRR antibody A-2 at a concentration of 10 mg/kg.

FuncS

Figure 8 is a graph showing that anti-Notchl NRR antibodies and gamma-secretase inhibitors reduce the viability of certain cancer cell lines.

Using anti-Notch1 NRR antibody A-2 and γ-secretase inhibitor N-[N-(3,5-difluorophenylacetyl)-L-alanyl]-S-phenylglycine tert-butyl ester (DAPT) and Compound E (CmpE). Cancer cell lines are shown on the x-axis.
