Description

Green fluorescent protein (GFP) is a 27kDa protein originally isolated from the jellyfish Aequoria victoria. It has endogenous fluorescent dye activity with a maximum excitation wavelength of 395nm and a maximum emission wavelength of 509nm, similar to fluorescein (1,2). The GFP gene was sequenced and the source of fluorescent dyes was discovered through the autocatalytic activity of certain amino acids (3,4). When it was discovered that the fluorescence developed rapidly when the protein was expressed and only molecular oxygen was needed and no other cofactors were needed, great interest in GFP was generated. Therefore, GFP can basically be expressed in fluorescent form in any prokaryotic or eukaryotic cell (5). GFP has been designed to produce a large number of different colored mutants, including blue, cyan, and yellow protein derivatives, BFP, CFP, and YFP (6-9).

Prot-r-AcGFP recombinant protein purified from E. coli. This epitope is located in the N-terminal 18 amino acids of the protein, the peptide MVSKGAELFTGIVPILIE, which can be found in Clontech and other GFP vectors
