Features

The red cAMP Maxensor-AVPR2 contains U2OS cells stably expressing the red cAMP Maxensor biosensor and vasopressin receptor 2 (unlabeled). Creative Biolabs' red cAMP Maxensor-AVPR2 cell line is designed to test compounds or analyze their ability to modulate vasopressin receptor 2. When the agonist binds to AVPR2, the G protein is activated, which in turn triggers a cellular response mediated by cAMP. This cell line has been verified by measuring the increase of cAMP in the cytosol and analyzing the distribution of the red cAMP Maxensor biosensor in the cell. This cell line allows image analysis of compound-induced stimuli. The use of [Arg8]-vasopressin as an agonist validates this highly reproducible assay in high-content analysis (HCA) and high-throughput analysis (HTA).

About Red cAMP Maxensor Biosensor:
The red cAMP Maxensor biosensor is a fluorescent peptide that changes its intracellular location in the presence or absence of cAMP. Before stimulating cAMP production, the fluorescent biosensor is located in the cell membrane. The increase in the concentration of the second messenger leads to changes in the structural folding of the red cAMP Maxensor biosensor, which promotes the relocation of cells in the transport of cell vesicles. In cell lines that co-express the red cAMP Maxensor biosensor and the GPCR of interest, the activity of living cells can be easily quantified by fluorescence granularity image analysis or fluorescence intensity analysis.

Assay Description

The arginine vasopressin 2 receptor belongs to the same subfamily of AVPR1A, AVPR1B, and OXT receptor G protein-coupled receptors. The activity of arginine vasopressin receptor 2 (AVPR2) mediated by Gs stimulates the second messenger system of adenylate cyclase. Creative Biolabs' V2 Vasopressin Receptor Assay allows the use of Maxensor biosensor technology to measure AVPR2 receptor activation. V2 vasopressin receptor signaling occurs through adenylate cyclase stimulation through the interaction of trimerized Gs protein, increased cAMP, and downstream L-type calcium channels. The cAMP-Maxensor biosensor measures receptor activation through cAMP accumulation.
