Description

Neurofilaments (NF) are approximately 10 nanometer intermediate filaments found in neurons. They are the main component of neuron cytoskeleton, mainly used to provide structural support for axons and adjust the diameter of axons. There are three main NF subunits, and the names of these subunits are based on the apparent molecular weight of mammalian subunits on SDS-PAGE. The light or minimum NF (NF-L) operating speed is 68-70 kD. The operating frequency of medium or medium NF (NF-M) is about 145-160 kD, and the operating frequency of heavy or highest NF (NF-H) is about 200-220 kD. However, due to the highly charged C-terminal region of the molecule, the actual molecular weight of these proteins is significantly lower. The level of NF gene expression is related to the axon diameter, which controls the speed of electrical signals propagating along the axon. Mutant mice with NF abnormalities have a phenotype similar to amyotrophic lateral sclerosis. NF immunostaining is common in diagnostic neuropathology. It is useful to distinguish between glial cells (positive for NF) and glial cells (negative for NF).
