Description

Methyl-CpG binding protein 2 (MeCP2) is widely expressed in tissues, especially in neurons. It is a nuclear protein that has been found to bind to methylated cytosines on CpG islands in DNA, but may also bind to hydroxymethylcytosine residues (1-3). Methyl and hydroxymethyl cytosine are usually found in DNA regions that regulate the expression of specific genes, and MeCP2 is considered a gene regulator, although it may have other functions. The MeCP2 protein contains a methyl-CpG binding domain and two AT hook domains, which are highly basic peptides that bind AT-rich DNA sequences. The MeCP2 gene MECP2 is located on the X chromosome. Loss-of-function mutations of this gene can cause Rett syndrome, which is a relatively rare neurodevelopmental and autistic disease that occurs almost exclusively in women, because similar mutations in MECP2 are almost always in men.
