Features

The cAMP Maxensor TSHR contains HEK293 cells stably expressing cAMP Maxensor-FP650 biosensor and thyroid stimulating hormone receptor (without label). The Creative Biolabs cAMP Maxensor TSHR cell line is designed to test compounds or analyze their ability to modulate thyroid-stimulating hormone receptors. When the agonist binds to TSHR, it activates the G protein, which in turn triggers a cellular response mediated by cAMP. This cell line has been verified by analyzing the intracellular distribution of cAMP Maxensor biosensor by measuring the increase of cAMP in the cytosol. This cell line allows image analysis of compound-induced stimuli. The use of thyroid-stimulating hormone as an agonist validates this highly reproducible assay in high-content analysis (HCA) and high-throughput analysis (HTA).

About Red cAMP Maxensor Biosensor:
The red cAMP Maxensor biosensor is a fluorescent peptide that changes its intracellular location in the presence or absence of cAMP. Before stimulating cAMP production, the fluorescent biosensor is located in the cell membrane. The increase in the concentration of the second messenger leads to changes in the structural folding of the red cAMP Maxensor biosensor, which promotes the relocation of cells in the transport of cell vesicles. In cell lines that co-express the red cAMP Maxensor biosensor and the GPCR of interest, the activity of living cells can be easily quantified by fluorescence granularity image analysis or fluorescence intensity analysis.

Assay Description

Thyroid stimulating hormone (also called thyroid stimulating hormone, thyroid stimulating hormone, or TSH) is a pituitary hormone that stimulates the thyroid. To analyze the activity of rhTSH, we used a stable cell line that stably expresses the thyroid-stimulating hormone receptor (TSHR). This cell line also expresses the cAMP-Maxensor biosensor, which increases its fluorescence intensity after TSHR is activated. When rhTSH binds to TSHR, it activates the G protein, which in turn triggers a cellular response mediated by the second messenger (cAMP). The cAMP-Maxensor TSHR cell line measures the increase of cAMP in the cytoplasm in a dose-response manner in the presence of rhTSH.
