Product Overview

Description

Cell line information: GLP1R cAMP Maxensor cell line
Official Full Name: Glucagon-like peptide 1 receptor
DNA Accession Number: NM_002062.4
Host Cell: U2OS
Resistance: G418 + Puromycin
Quantity:> 3 x 10^6 cells / vial
Storage: Liquid Nitrogen

Features

The red cAMP Maxensor-GLP1R contains U2OS cells stably expressing the red cAMP Maxensor biosensor and glucagon-like peptide 1 receptor (without label). Creative Biolabs' red cAMP Maxensor-GLP1R cell line is designed to test compounds or analyze their ability to modulate glucagon-like peptide 1 receptors. When the agonist binds to GLP1R, it activates the G protein, which in turn triggers a cellular response mediated by cAMP. This cell line has been verified by measuring the increase of cAMP in the cytosol and analyzing the distribution of the red cAMP Maxensor biosensor in the cell. This cell line allows image analysis of compound-induced stimuli. The use of human GLP-1 as an agonist in high-content analysis (HCA) and high-throughput analysis (HTA) validates this highly reproducible assay.

Application Notes

About Red cAMP Maxensor Biosensor:
The red cAMP Maxensor biosensor is a fluorescent peptide that changes its intracellular location in the presence or absence of cAMP. Before stimulating cAMP production, the fluorescent biosensor is located in the cell membrane. The increase in the concentration of the second messenger leads to changes in the structural folding of the red cAMP Maxensor biosensor, which promotes the relocation of cells in the transport of cell vesicles. In cell lines that co-express the red cAMP Maxensor biosensor and the GPCR of interest, the activity of living cells can be easily quantified by fluorescence granularity image analysis or fluorescence intensity analysis.

Assay

GLP-1 Glucagon Receptor Assay

Assay Description

GLP-1 glucagon receptor; also known as GLP1R; is a receptor protein that exists in beta cells of the pancreas and brain neurons. Creative Biolabs' GLP-1 glucagon receptor test allows the use of Maxensor Measure GLP1R receptor activation biosensor technology. GLP-1 glucagon receptor signal is stimulated by adenylate cyclase through trimeric Gs protein, increase cAMP and downstream L-type calcium channel interaction. cAMP-Maxensor biosensor Receptor activation is measured by cAMP accumulation.

The high reproducibility of this assay allows monitoring of the GLP-1 glucagon receptor activation process in high-throughput screening.

Mycoplasma Testing

The cell line has been screened using the luciferase based mycoplasma detection kit to confirm the absence of mycoplasma species.

Sterility Testing

Sterility testing was performed in accordance with USP and EP regulations. All of our sterility testing is performed in an isolator or clean room environments. The cell line has been screened using the membrane filtration testing methods to confirm the absence of aerobic, anaerobic and fungi microorganisms.

Genetic Stability Testing

Cell genetic stability study was perfomed under ICH guidelines. We provide guidance on the appropriate testing program upon your requirements.
