Description

Lentiviral vectors based on human immunodeficiency virus 1 (HIV-1) have become the most commonly used vectors for gene transfer research. The dominant feature of lentiviral vectors is the ability of gene transfer and integration into dividing and non-divi. The pseudotype envelope with vesicular stomatitis virus envelope G (VSV-G) protein expands the range of target cells. Lentiviral vectors have been shown to deliver genes to neurons, lymphocytes, and macrophages, while previous retroviral vectors cannot use this cell type. Lentiviral vectors have also been shown to effectively transduce brain, liver, muscle and retina in vivo without toxicity or immune response. Recently, lentivirus systems have been widely used to effectively integrate siRNA into various cell lines and primary cells in vitro and in vivo.

Lentiviral particles are produced from 293T cells by transient transfection of plasmids encoding viral particle components. For safety considerations regarding HIV-1 infectivity, recent lentiviral packaging systems have divided the viral components into 3 or 4 plasmids.

The pLenti-EF1-MCS + -PGK-PURO lentiviral expression vector contains multiple cloning sites (MCS) downstream of the EF1α promoter and a puromycin resistance gene driven by the PGK promoter. Lentiviral expression vectors contain multiple restriction sites, making it easy to clone the promoter of your choice and the gene of interest.
